Sophie Thatcher was born in Chicago, Illinois to a Mormon family, but she has since left the church. She has three siblings, including an identical twin sister named Ellie. Sophie describes her family as being musically inclined. She sang in her church choir while her mother played the piano. She began acting at the age of four and went to a performing arts school for musical theatre.

Sophie’s on-screen debut was a guest role in 2016 on the police procedural series Chicago P.D. That same year, she guest-starred on two episodes of Fox’s supernatural horror series The Exorcist.

Sophie made her feature film debut in Prospect (2018), with Pedro Pascal and Jay Duplass. Sophie played the lead role of Cee, who travels with her father (Duplass) to an alien moon to mine for gems in the moon's poisonous forest. The film received positive reviews, and Sophie was commended for her performance.

In the years following, Sophie established her career on television. She was a regular on the 2020 series When the Streetlights Go On, playing Becky Monroe, and had a supporting role on the 2022 limited series The Book of Boba Fett, in the role of Drash.

In 2021 Sophie appeared as Natalie on Yellowjackets. In a show that has two parallel storylines, one set in the past, the other in the present, Sophie shared the character of Natalie with Juliette Lewis, who played the adult version of the character.

In 2024, Sophie appeared in Heretic (2024) starring Hugh Grant. She played Sister Barnes, who is a member of a Mormon missionary and tries to convert others to Mormonism. Sophie said playing the character was a very personal experience as it allowed her to reflect on the time when she was a part of the church.

In 2025, Sophie starred in Companion (2025) as Iris, a sentient sex robot who seeks revenge. The film is directed by Drew Hancock and also stars Jack Quaid.
