Born in Oxford, Joseph Mawle grew up on a farm with his parents and two siblings. At a young age, Joseph was diagnosed as severely dyslexic and from the ages of 13 to 16 he went to a boarding school for special needs. He left the school to become an actor, but contracted a virus that left him partially deaf. He acted with the Box Clever Theatre Company in London and eventually won a scholarship to Bristol Old Vic Theatre School. When Idol Magazine asked him about his idol, Mawle said, "I went to train at the Bristol Old Vic specifically because Daniel Day-Lewis has trained there."

His first major role was in the 2003 stage production of Shakespeare's Troilus and Cressida, in which he played Troilus. In 2006, Joseph appeared in the TV movie Soundproof when a deaf actor was needed for the lead role. For his performance, he was nominated for the RTS Breakthrough On Screen Award. He also starred in the British TV productions Dunkirk, Persuasion and Clapham Junction.

In 2008, Joseph returned to theater to play the title role in The Last Days of Judas Iscariot. He played regular roles on television series such as Waking the Dead, Five Daughters and Game of Thrones.

The English actor was in the TV movie Dive, then played alongside Dominic Cooper in Freefall. He rejoined Cooper in 2012's Abraham Lincoln: Vampire Hunter, playing Thomas Lincoln. He also played a role in The Awakening (2011) with Rebecca Hall, and stars alongside Henry Cavill and Bruce Willis in The Cold Light of Day (2012).
