While studying at Boston College, Massachusetts native Amy Poehler became involved with sketch comedy when she joined a troupe called My Mother's Flea Bag. She moved to Chicago to study at Second City in 1993, then joined the Upright Citizen's Brigade comedy group in 1996. She made her TV debut the same year, providing her voice and writing talents to the TV movie Escape From It's a Wonderful Life. She also played Andy Richter's little sister Stacy on Late Night with Conan O'Brien.

In 1998, Upright Citizen's Brigade moved to New York, where they got their own sketch comedy series on Comedy Central, which lasted three seasons. Meanwhile, Amy made her film debut in Tomorrow Night (1998), which screened at the Sundance Film Festival. She also made a guest appearance on the sitcom Spin City starring Michael J. Fox. Her major motion picture debut came with a supporting role in Deuce Bigalow, Male Gigolo (1999), starring Rob Schneider. She then appeared in the box office flop Wet Hot American Summer (2001) and The Devil and Daniel Webster (2001), which was never theatrically released in America.

Amy joined Saturday Night Live as a featured player in 2001 and was upped to repertory cast in January 2002. In 2004, she had the distinction of appearing in two major motion pictures opening on the same day—Envy, starring Jack Black and Ben Stiller, and Mean Girls, starring Lindsay Lohan and scripted by her good friend, SNL writer/performer Tina Fey.

Amy lent her voice to two films, Alvin and the Chipmunks: Chipwrecked (2011) and The Secret World of Arrietty (2012). She then starred in They Came Together (2014), Are You Here (2014) also in the comedy Sisters (2015) alongside Tina Fey.

Amy married actor Will Arnett in 2003. He starred on the critically-acclaimed Fox sitcom Arrested Development. Amy has made several guest appearances on the show, playing his wife. She also starred as Leslie Knope in the television series Parks and Recreation.

Unfortunately, her marriage to Arnett ended in divorce in 2016. They share custody of their two children. Her memoir, Yes, Please, was published in 2014.
